However, the role of metabotropic GABA(B) receptors in depression is not well defined. Effects of acute and chronic treatment with amitryptyline on the sleep-wake activity of rats.Amitriptyline ( Elavil ) (1, 5 or 15 mg/kg intraperitoneally, twice a day) was administered to rats and the sleep-wake activity was recorded for either 24 hr (1 mg/kg) or 12 hr (5 or 15 mg/kg) on the day before treatment with Amitriptyline ( Elavil ), on days 1 and 5 of the treatment and on day 6, when the drug was withdrawn. 5-Hydroxytryptamine depletion (>90%) online medicines hydrochlorothiazide female generic isordil abolished the antidepressant-like behavior of CGP56433A (10 mg/kg) by attenuating the increase in swimming. We demonstrate that the selective GABA(B) receptor antagonists CGP56433A [[3- 1-(S)-[ 3-cyclohexylmethyl)hydroxy phosphinyl -2-(S) hydroxy propyl]amino aloise]benzoic acid; 1-10 mg/kg] and [3-[[1-(S)-3-dichlorophenyl)ethyl]amino]-2-(S)-hydroxy-propyl]phenylmethyl-phosphinic cetirizine hcl acid hydrochloride; 3-10 mg/kg] had a similar profile to the selective serotonin reuptake inhibitor Fluoxetine ( Prozac ); they decreased immobility and increased swimming behavior. The results show that the increase in non-REM sleep is as characteristic of Amitriptyline cetirizine hcl ( Elavil ) as the reduction of REM sleep, and that these effects are resistant to chronic treatment when the dose is small. In the first 3 hr Amitriptyline ( Elavil ) increased non-REM sleep (NREMS), and decreased REM sleep (REMS) and wakefulness; the effects were dose-dependent. To further assess the possible interaction between GABA(B) receptor antagonism and serotonin, rats were pretreated with the tryptophan hydroxylase inhibitor para-chlorophenylalanine. In contrast, the novel GABA(B) receptor-positive modulator GS39783 (10-40 mg/kg) did not display antidepressant-like activity in the modified forced swim test. Amitriptyline ( Elavil ) at 1 mg/kg had no effect on the sleep-wake activity during the dark period. To assess the role of GABA(B) receptors in antidepressant-related behavior, we examined a number of selective GABA(B) receptor ligands (novel positive modulators and antagonists) on behavior in the modified forced swim test. The changes in non-REM sleep and wakefulness (W) were follo by a compensatory reaction 6-12 hr after the treatment. The effects of chronic injection of Amitriptyline ( Elavil ) on non-REM sleep revealed a definite decrease only in the huntlee of the 15 mg/kg dose. Together, these data demonstrate that GABA(B) receptor antagonists via an interaction with the serotonergic system display antidepressant-like properties and therefore represent a novel approach for the treatment of depression.. Rebound of REM sleep appeared after withdrawal of the 5 and 15 mg/kg doses. The modified forced swim test has recently emerged as an excellent tool to assess behaviorally the role of monoamines in antidepressant action.
